In 1903, 21-year old William S. Harley and 20-year old Arthur Davidson the first production Harley-Davidson motorcycle that was hand-built in a small garage in Milwaukee. Today, Harley-Davidson, Inc. employs more than 8,200 people and has 1,110 dealership worldwide. Its corporate headquarters are still located in Milwaukee, Wisconsin. The domestic production facilities are in the Milwaukee area, East Troy and Tomahawk, Wisconsin; York, Pennsylvania; and Kansas City, Missouri. Design, customer focus and sales have placed this company is in a very unique and desirable marketing situation. Design is critical to the continued success of this company. For some companies, the word design may bring thoughts of sweeping changes and new model lines. However, for the design team at Harley-Davidson, it inspires thoughts of being faithful to the classic lines and sounds of a Harley-Davidson motorcycle. This is not to say that the designers are not allowed and encouraged to improve on the hugely successful line of motorcycles, just the opposite is true. Ken Schmidt, director of communications for Harley-Davidson, stated that: â€Å"We are constantly improving and modernizing the machine, yet not every component. That is what our customers want, and that’s also, I believe, what sparks the strong emotional attachments that Harleys generate.† In the early 1980’s, the company was plagued with quality control and reliability issues that nearly caused the company to go out of business. At this point, it was up to the design and quality control engineers to revamp the mechanical and electrical portions while staying true to the classic line of the motorcycle. They succeeded in resolving the problems and saved the company. William G. Davidson, who heads up a team of five product designers, is quick to add, â€Å"It’s important to understand that we’re not in the business of making antiques. Every year we make further improvements to our engines and chassis. But from a styling standpoint, we have to incorporate improvements without compromising `the look.’ If we move a bolt or re-route one hose, our customers take note and call us on it. A Harley isn’t shrouded in fiberglass like so many other bikes. Everything we do is right out in the open.† This mindset prevents lifelong riders from feeling  alienated and accusing the company of â€Å"selling out† in the pursuit of increased revenue and profits. Harley-Davidson understands that it is as important to maintain their current enthusiasts as it is to develop new ones. No where is this more evident than in their mission statement: â€Å"We fulfill dreams through the experience of motorcycling, by providing to motorcyclists and to the general public an expanding line of motorcycles and branded products and services in selected market segments.† Customer focus is another area of marketing where this company excels. Factory sponsored rallies and test drives are just two ways that Harley stays in touch with their customers. The largest sponsored rally occurs each year at Sturgis in the Black Hills of South Dakota. This event draws 400,000 riders from around the world. There are hundreds of company employees there as well, not only to enjoy the rally, but also to gather information from the enthusiasts. There is no better or more efficient way to survey your customers than to speak with them directly after they just traveled to an event on your product. This â€Å"close-to-the-customer† philosophy allows the company to use these rallies as product development centers. It allows the company leadership to get instant responses on both current products as well as new products that may soon go into production. Customers respond positively to this type of product survey, because they feel like their opinion matters and will influence the next year’s product line. Another marketing tool that has been quite successful for this company has been the factory-sponsored test-drives. The factory sends a tractor-trailer filled with demo bikes to each dealership that is willing to participate. All of the new models are available and anyone with a helmet and a valid license to operate a motorcycle can try them out. For many of the dealerships, it is the busiest day of the year. Harley-Davidson headquarters employs eight merchandising specialists that develop â€Å"designer stores†. These stores are much different from the standard â€Å"garage-type† store of the past. These stores are designed to draw customers in, surround them with motorcycles, and provide an inviting retail environment. Dealerships that have converted their shops to the Designer Store concept have seen soaring revenues and rapid return on investments.  Inventive marketing strategies have kept Harley-Davidson as the leader of America’s big-bike market. It is not just the motorcycles that are in high demand. Harley has developed a very profitable retail merchandising line. This line includes clothing, tattoo patches, coffee mugs, belt buckles, infant wear and memorabilia. Most would think that this company must spend a significant portion of their revenue on advertising. The opposite is true, in 1995 the company generated $1.3 billion in revenue while spending less than $2 million on advertising costs. â€Å"Word of mouth† is their main form of advertising. Company executives feel that their customers are their sales force, after all, satisfied customers are the better than advertising. Harley-Davidson is in a unique marketing situation because they are not selling transportation; they are selling a lifestyle. Customers routinely wait between six and eighteen months for delivery of a new model. That type of product loyalty is hard to find and not easily developed. The waiting list for a new Harley-Davidson is now more than a year, and used bikes are more expensive today than when they were new. How can this possibly be true when new cars depreciate quicker than you can make the payment? The reason is the supply-to-demand ratio and Harley-Davidson has mastered this ratio. Although they produced more than 300,000 bikes last year, demand is still on their side. Many new customers look at purchasing a Harley as an investment that you can enjoy on a daily basis. The first turning point for the company was when they changed the engines back in 1984. The second was when the â€Å"baby boomers† hit their mid-life crisis. Since then, dealers have struggled to keep display models on the floor. Scarcity allows the company to achieve higher profitability, and it also helps protect the investment for previous buyers. Women are the last great frontier for this company and they are beginning to focus more on that segment of the market. They have begun to place advertisements in women’s magazines in an attempt to change the company’s image to a more family-oriented one. They have also begun to make engineering changes to some of the models. Some of the new bikes will have  smaller handlebar grips, an easier pulling clutch, and lower seats. â€Å"Riders Edge† is a program at Harley dealerships providing riders’ education classes to help novices learn to ride and get licensed. Technology upgrades such as liquid-cooled, high-powered, engines and lightweight materials will make bikes more powerful, but also less difficult to handle. All of these changes are being implemented to encourage women to purchase their own bikes as opposed to simply being passengers. Customers can sense that the Harley-Davidson employees care about them and their concerns. The company takes this responsibility so seriously that they developed a list of â€Å"core values†. These are their values. They are the heart of how we run their business. They guide their actions and serve as the framework for the decisions and contributions their employees make at every level of the company. Tell the truth, be fair, keep your promises, respect the individual, and encourage intellectual curiosity. Because of their supply-and-demand ratio, it would be very easy for this company to develop a â€Å"take it or leave it† attitude with their customers. The video persuades you that the types of individuals that drink this beer are ones who care and respect others. Individuals that believe that The choices we make reveal the true nature of our character. The introduction is the most important feature of this commercial. Because there is no dialog, the visuals are what tell the story. The commercial begins with the song playing right on cue of the ball missing the goal. If you notice, the video is in slow motion at this point. When the ball comes down and is rebounded the camera is repositioned on the guys rolling down the court to symbolize how strenuous these guys are working. Their dedication Is displayed not only by the looks on their faces when the camera Is zoomed In, but by the sweat dripping aimlessly down their brows. Consequently, the announcer comes n and states dedication when one of the men fall down In the wheelchair trying to retrieve the ball but instantly gets back up to get back in the game This is not only emotionally appealing to the audience because they are playing the diligent game of basketball, but because these guys have to work even harder due to the fact that they are paraplegics.The final goal is made and all but one of the guys gets up from their wheelchair which visualizes the devotion these guys demonstrate to the one guy who actually is handicap. Once again this an attempt from the creators of this commercial to emotionally appeal to its audience by representing the loyalty of playing basketball in the most unbiased way possible to a paraplegic friend. The music In the background plays an important role In this commercial. The Cinematic Orchestra To Build a Home Is very Inspirational to the visuals to this commercial.The tempo perfectly aligns with the events In this story line. It begins with this slow tempo to signify the sadness in the events but then climaxes when the goal is made ND shows that not everyone in this video is actually in a wheelchair and it finalizes when the tempo becomes lowered and constant symbolizing a calm and collectivities that we are all the same despite our differences. The other category is what Feldman (2009) refers to as background stressors and are the things that impact our daily life. South Florida has many factors that contribute to our stress level, high unemployment rate 12.1% , high number of families living in poverty approximately 16%, (U.S. Census Bureau, American Fact Finder at (http://factfinder.census.gov), a housing crisis that has contributed to many residents losing their homes, overcrowded roads, high crime rate, and during hurricane season the weather. When we compare ourselves with people living in smaller cities, villages, other areas of the country or even other countries, or those whose socio economic status allows them to maintain their life style during difficult economic times we could probably see a difference in the stress level. Who has not stressed over driving in the Palmetto or I-95. Who has not stressed over the evening news or the possibility of not having a job in very difficult economic times? Who has not stressed over the possibility of being affected by the housing crisis? Who does not stress over a situation at work over which we have no control? These are stressors we live with and deal with on a daily basis. Work Stress and Mental Health Some people have higher levels of stress because they might have a very stressful job. Police officers, fire fighters, soldiers in a war zone, health care providers, long distance truck drivers, and yes, even educators (think of a classroom full of students where you are responsible for making sure that they are learning the skills they need to succeed) have very stressful jobs. According to Elkin and Rosch (1990) workplace related stress in a major problem in the U.S. and it creates a major expense for corporations. Koeske, Kirk, and Koeske (1993) indicate that all jobs have some level of stress but jobs that are in the human services have additional stressors because they â€Å"derive from intense involvement in the lives of others†. (p.319). They also refer to the type of stress experienced by individuals working in human services as â€Å"burnout† (Koeske, Kirk, and Koeske, 1993, p.319). Another factor that can influence our stress level is how we handle lifeâ€℠¢s demands. There are different ways of reacting to the situations we face every day of our lives. We have all heard of expressions such as ‘road rage’ and ‘going postal’. These are terms that we have come to identified with violent reactions to stressors caused by situations all of us face every day. How many of us have experiences in the road that would cause us to display behavior that is outside of what we consider to be within a normal range? How many of us have had experiences at work with a supervisor or a coworker where we have had to use all of our self control not to respond in an appropriate manner? We all have had these experiences and how we handle them depends on different factors such as our own characteristics, or external factors that could be impacting us at any particular moment. Symptoms of stress and anxiety caused by work According to Tetrick and LaRocco (1987) â€Å" work conditions can lead to perception of stress. Perceived stress, in turn, leads to job-related strains such as dissatisfaction, boredom, and turnover, and to individual strains such as anxiety, depression, and physical illness† (p.538). There are other factors that can cause high level of stress in a job for example high demands on the part of an employer, (i.e. constant deadlines, extended work hours) and low control on the part of the employee (i.e. very little control over the day to day activities in the workplace) and low rewards for effort can increase the chances of having serious health problems such as anxiety, use of prescription or over the counter drugs to deal with stress. (Workplace Mental Health Promotion, 2010). According to Tetrick and Larocco (1987) in addition to these external factors related to our jobs and the type of work conditions we have we also have internal factors that they call â€Å"internal char acteristics (i.e., personal characteristics) and external conditions (i.e., situational characteristics)† (p. 358). Pachner (2010) stated that In response to feelings of insecurity or ineffectiveness many people can avoid social interactions with colleagues and can develop physical symptoms such as headaches, insomnia, high blood pressure and even heart attacks. Many of us have experiences these types of feelings or reactions when we have found ourselves in stressful jobs. These health related issues can impact the individual, his/her family, the workplace environment and the community in general. Higher costs for health care, incidents of road rage or workplace violence, as well as in some instances domestic violence can all be linked to stressed caused by work. Individuals at risk of experiencing conditions of stress and anxiety As indicated before most of us are at risk of experiencing stress and a burnout feeling at any time. Those of us who are in the fields where we deal with people or a daily basis or where high demands and where we have little control of our work environment will be more impacted than others. Additionally as mentioned before certain type of personality characteristics will increase or levels of stress. Some of the internal characteristics might be our way of handling difficult situations, our need to control our environment, our need to overachieve, even our need to please others. Some individuals have a need to have control over all aspects of their lives and when decisions are made by others and they have no control over these decisions it impacts their well being. Stressful jobs such as those involving high risk or oneself, i.e. law enforcement, military duty in a war zone, can cause higher level of stress. Jobs where demands are placed in our abilities to deal with others can also create stress, i.e., healthcare professionals, customer service personnel, teachers, and other professionals who deal directly with others. The stress caused by these types of jobs can influence our health, our attitudes, and even our home life. In many instances individuals do not have the opportunity to decompress before arriving home to deal with the other stressors that we all confront. When we leave our jobs we then need to face the different challenges surrounding us in our community i.e. traffic, weather, etc. Dealing with stress and anxiety According to Elkin and Rosch (1990) To prevent burnout individuals need to monitor their mental and physical health and maintain a healthy lifestyle that includes a healthy diet, leisure time and connecting with family and friends. All of us need to understand the importance of maintaining a healthy equilibrium between work and our own personal life. Giving in to the demands of a stressful job can only serve to create additional health problems such as heart conditions, high blood pressure, dependency on over the counter, illegal, or prescription drugs. It will also contribute to other problems such as lack of sleep, isolation, and sometimes depression. Perhaps we need to look at how other societies live their daily lives. For example traveling in the Mediterranean, specially the Greek islands, can give you a different perspective of how life can be. The Greek Islands have a low rate of heart disease and having traveled there myself I can see how their lifestyle contributes to that l ow risk. They are very active and involved in activities that promote healthy lifestyles i.e. fishing. They eat healthy and most of the time is food that is prepared in a healthy way. They take time to rest during the day and they take the time to enjoy family and friends and establish social connections. We, on the other hand, because of our work schedules, the time it takes us to get to and from work, the demands we place on ourselves, rarely have time to socialize and spend time with family or friends. Perhaps corporations should take into consideration the need to provide time for employees to establish relationships at work that are not just work related but that go beyond the routine of the workplace. Encouraging employees to have an active life style would also help the stress since physical activity reduces stress level. Having a good health insurance that promotes healthy life style such as by encouraging preventing medicine and membership in health clubs will contribute to reduce the stress level. We also need to take proactive steps to reduce the stress level caused by our jobs. One of the steps we can take is to accept that our personality can play a major role on how we adapt and deal with the job demands and take steps to change the extra stress we create for ourselves. If we like to control what we do at all times and like to plan ahead for all events but work in a job that is constantly changing we can begin by accepting that we can control the time we leave for work, what we take for lunch, and what route we take to get to and from work and that will give us a sense of control for at least part of the day. If we like to achieve perfection or please those around us we can begin by establishing realistic goals that we can achieve every day rather than trying to be the best in all possible aspects. Understanding ourselves, our needs, our personality, and the demands of our jobs can help us to decrease the level of stress we experience. There are factors over which we have no control i.e. the crime rate in our area, the weather, the overcrowded highways, etc. However we can certainly control other aspects of our lives such as how we handle the demands of our job, the type of lifestyle we choose to live and the amount of quality time we choose to spend with family and friends who are important to us. Knowledge and Career Understanding that there is an entire group of problems that are known as â€Å"psychophysilogical disorders that often result or are worsened by stress† (Feldman, 2009, p.421) can help us to realize that these are very real medical problems that are â€Å"influenced by an interaction of psychological, emotional, and physical difficulties† Feldman, 2009, p.421). Having gained some basic knowledge of the different aspects of what can cause stress in our lives such as work, environment, social demands, etc. can help us manage our lives and careers in a way that will help us to achieve our goals, maintain healthy relationships, succeed at our jobs, and maintain a healthy life style. According to Feldman (2009) our relationship with others can help us deal with our stress. The social support we can establish can help us to lower our stress level and the support we show others can help them as well. Coping with the daily stressors we face, and doing so as a member of a group of family, friends, coworkers, or neighbors can help us to maintain a healthy level of stress and will certainly reduce the risk of health problems associated with major stress in our life. This will in turn help us to be successful in our careers because we will achieve a healthy balance of work goals, family goals, community commitment, and a sense of well being.

Another difference between these two types of food is the cost. Canned foods are much more expensive than the fresh foods. Which Antibiotic is the Most Effective on Bacteria? INTRODUCTION Antibiotics The word antibiotic comes from the Greek anti meaning against and bios meaning life (a bacterium is a life form). Antibiotics are also known as antibacterials, and they are drugs used to treat infections caused by bacteria. Bacteria are tiny organisms that can sometimes cause illness to humans and animals. The singular word for bacteria is bacterium. Such illnesses as tuberculosis, salmonella, syphilis and some forms of meningitis are caused by bacteria. Some bacteria are not harmful, while others are good for us. Before bacteria can multiply and cause symptoms our immune system can usually destroy them. We have special white blood cells that attack harmful bacteria. Even if symptoms do occur, our immune system can usually cope and fight off the infection. There are occasions, however, when it is all too much and our bodies need some help from antibiotics. The first antibiotic was penicillin. Such penicillin-related antibiotics as ampicillin, amoxicillin and benzylpenicilllin are widely used today to treat a variety of infections these antibiotics have been around for a long time. There are several different types of modern antibiotics and they are only available with a doctors prescription in industrialized countries. How do antibiotics work? Although there are a number of different types of antibiotic they all work in one of two ways: A bactericidal antibiotic kills the bacteria. Penicillin is a bactericidal. A bactericidal usually either interferes with the formation of the bacteriums cell wall or its cell contents. A bacteriostatic stops bacteria from multiplying. Uses of antibiotics An antibiotic is given for the treatment of an infection caused by bacteria. They target only bacteria they do not attack other organisms, such as fungi or viruses. If you have an infection it is important to know whether it is caused by bacteria, and not a virus or fungus. Most upper respiratory tract infections, such as the common cold and sore throats are generally caused by viruses antibiotics do not work against viruses. Antibiotics are overused or used incorrectly there is a chance that the bacteria will become resistant the antibiotic becomes less effective against that type of bacterium. A broad-spectrum antibiotic can be used to treat a wide range of infections. A narrow-spectrum antibiotic is only effective against a few types of bacteria. There are antibiotics that attack aerobic bacteria, while others work against anaerobic bacteria. Aerobic bacteria need oxygen, while anaerobic bacteria dont. Antibiotics may be given beforehand, to prevent infection, as might be the case before surgery. This is called prophylactic use of antibiotics. They are commonly used before bowel and orthopedic surgery. Bacteria The word bacteria is the plural of bacterium. Grammatically the headline should just say What are bacteria? Bacteria are tiny living beings (microorganisms) they are neither plants nor animals they belong to a group all by themselves. Bacteria are tiny single-cell microorganisms, usually a few micrometers in length that normally exist together in millions. A gram of soil typically contains about 40 million bacterial cells. A milliliter of fresh water usually holds about one million bacterial cells. Planet Earth is estimated to hold at least 5 nonillion bacteria. Scientists say that much of Earths biomass is made up of bacteria. 5 nonillion = 5,000,000,000,000,000,000,000,000,000,000 (or 51030) (Nonillion = 30 zeros in USA English. In British English it equals 54 zeros. This text uses the American meaning) Bacterial cell walls Bacterial cell walls consist of layer of peptidoglycan which is made up of many parallel polysaccharide chains with short peptide cross-linkage forming an enormous molecule with net-like structure. However, there are two different types of bacterial cell wall, which can be distinguished by Gram staining, a staining technique developed by Christian Gram in 1984 and still in use today. Before staining, bacteria are colorless. The cell walls of Gram-positive bacteria have a thick layer of peptidoglycan containing chemicals such as teichoic acid within their net-like structure. The crystal violet in the stain binds to the teichoic acid and resists decolouring in the rest of the process, leaving the positive purple/blue color. The cell walls of Gram-negative bacteria have a thinner layer of peptidoglycan with ni teichoic acid between the two layers of membranes and then an outer membrane-like layer made up of lipopolisaccharides. Any crystal violet which does not bind is readily decolourised and replaced with red safranine in the Gram stain. So cells appear red. PROBLEM STATEMENT Which antibiotic is the most effective on bacteria? APPARATUS 200 ml of micropipette, conical flask, forceps, 100 ml beaker MATERIALS Petri dish, sample of E.coli and staphylococcus, 3 different types of antibiotic which are carbenicilin, streptomycin and tetracycline, distilled water, Dettol handwash, 75 % of ethanol, agar, tissue paper VARIABLES Fixed variable : volume of bacteria Manipulated variable : types of antibiotic Responding variable : area of inhibition zone HYPOTHESIS The most effective antibiotic to inhibit bacteria growth is ampicillin PROCEDURE First of all, wash out hands with the soap or handwash. The working area is sprayed thoroughly with the disinfectant spray. It is left for at least 10 minutes, and wiped with a paper towel. An agar plate seeded with is prepared. The petri dish is labeled on the base at the edge out of name, the date and type of bacterium it is inoculated with by usng permanent marker pen. After finishing marking the petri dish, the Esterichia coli bacteria is poured by using micropipette into the petri dish on the marked labeling and distribute it evenly. The agar is taken out from 60 ÃÅ'Ã…  C oven. The mouth of the conical flask containing the agar is warmed in the flame to prevent any different bacteria from surrounding grow inside it. The agar is left for 10 minutes for it to solidify before putting the sterile disc dip into three different antibiotics. The forceps are flamed and used them to pick up disc or Mast ring. It is dip into the antibiotic solution and is placed firmly in the centre of the agar. The dish is taped securely with two pieces of adhesive tape and kept it upside down at room temperature for 24 hours. Step 1 until steps 7 are repeated by using another type of bacteria which is staphylococcus. Our hands are washed with soap or handwash and the bench is cleaned again using the 75 % of ethanol. After the incubation, the plate should be looked at carefully but do not open it. Where bacteria have grown the plate will look opaque, but where the antibiotics have inhibited growth, clear zones called inhibition zones will be seen. The diameter of the inhibition zones is measured in milimetres and the information is used to decide which antibiotic is most effective at inhibiting the growth of the bacterium. The data is collected from other members of the class who used the other bacterial cultures. PRECAUTIONS When carrying out the experiment, we should work very closed to the Bunsen burner to prevent any impurities in each apparatus that is used. Do not seal completely the upper and lower part of petri dish. Both petri dishes contain different bacteria need to be inverted to prevent the water vapour from forming at the upper site of the agar. It might affected or overshadow the inhibition zones that are formed inside the dish. Make sure our hands are constantly being wash with disinfectant before touching any apparatus. Before using any apparatus, they should be sterile to prevent any unwanted impurities to grow in the petri dish. Do not open the petri dish after incubation to prevent from infection. Explanation of the data Table above shows the area of inhibition zone of the bacteria growth on two different bacteria. Four types of antibiotics are used to be investigated which is the most effective on a particular bacteria. They are tetracycline, streptomycin, carbenicillin and ampicillin. Whereas two types of bacteria are used which are staphylococcus and Esterichia Coli. For E.coli, ampicilin showed the greatest inhibition area of bacteria growth with 10.2 cmÂÂ ². Another antibiotic that showed the closest reading is streptomycin. Tetacyclin and carbenicilin showed 5.3 cmÂÂ ² and 4.9 cmÂÂ ² respectively. This result clearly shown that ampicilin is the most effective antibiotic to inhibit the growth of E.coli. Besides that, E.coli is a gram negative bacteria. The cell walls of Gram-negative bacteria have a thinner layer of peptidoglycan with no teichoic acid between the two layers of membranes and then an outer membrane-like layer made up of lipopolisaccharides. In addition to that, Staphylococcus also had a higher inhibition zone of bacteria on ampicilin. Other bacterias have no effect as great as ampicilin which can be consider as a strong antibiotic. Staphy is a gram positive bacteria. The cell walls of Gram-positive bacteria have a thick layer of peptidoglycan containing chemicals such as teichoic acid within their net-like structure. The crystal violet in the stain binds to the teichoic acid and resists decolouring in the rest of the process, leaving the positive purple/blue color. That is why ampicilin is said to be the most effective antibiotic on both bacterias. Limitations There are some limitations that cannot be avoided when carrying out the experiment. The first one was every apparatus that we used had been sterile by the laboratory assistant. Thus we had to wash our hands thoroughly before touching anything. We even could not talk during the preparation. The problem was we carried out the experiment on the labs table, instead of using the cupboard fume to maintain the sterile apparatus. Thus, there might be some of the apparatus that we were using had been contaminated. This condition might affect the reliability of the experiment hence the expected results might not get at the end of the experiment. Besides that, we had to wash our hands by using the dettol hand soap before we start and end the experiment. We had to remove all the impurities and bacteria and that was the purpose of washing hand. The problem rose as we did not know whether we had washed our hand to the maximum cleanliness. Consequently, all the apparatus and materials that we used might be affected by our hand. This is done so that there are no infections or contaminations on the agar that might give problems later. Sources of error There are some sources of error when carrying out this experiment. Firstly, we prepared the experiment at room temperature. It was supposed to carry out in sterile medium to prevent anything from affecting during the preparation of bacteria and antibiotics. Next, the major problem that could be seen during the experiment was most of the groups could not be able to solidify their agar to the maximum hardness. Consequently, when inverting the petri dish, all its contents would be messy inside the petri dish and the disc had mixed to one another. Thus, the antibiotics on each disc also had mixed that could affected the whole result. To overcome this problem, students should let the agar solidify before inverting the petri dish. Another problem was some of the groups do not invert their petri dish before keeping it inside the incubator. This would cause the water vapour to form at the upper part of the petri dish. It would also affect the growth of the bacteria in agar. Fourth, the layer of agar inside the petri dish was too thin that caused it fell to the bottom when inverting it. Besides ruining the shape of the agar and the position of the discs in the agar, the thin layer of agar also inhibits the growth of bacteria because it lacked of nutrients. To prevent this, the layer of agar should be half of the petri dish. Further work To get more reliable and accurate result, this experiment should be repeated by varying the temperature to investigate the activity of antibiotic on bacteria. This means that increase the temperature would cause the antibiotic to act more rapidly compared to the temperature that we used before. Thus we do not have to wait for 24 hours to see the inhibition zone. This manipulated variable also do not waste the time consuming for the experiment. Moreover, as the antibiotic shows more faster effect, thus the bacteria and antibiotic should be put in separately different petri dish to prevent it from overlapping. CONCLUSION The most effective antibiotic is ampicilin. The hypothesis is aceepted.
